In Majikoi - Oh! Samurai Girls! ist die Stadt Kawakami für seine früheren Samurais berühmt. Yamato, ein Schüler im zweiten Schuljahr in der Kawakami Highschool, verbringt viel Zeit bei seinen Freunden. Sie kennen sich schon seit ihrer Kindheit und haben viele Dinge gemeinsam gemacht.
